You can use those tags to display ads or affiliates offers. You can enter the code or call an external PHP functions.

For the adblocks, you can use the code that's in the readme.txt from the default themes. You will need to define the size for each of them. Here are an example:

[adblock1] could be 250x250

[adblock2] could be 468x60

[adblock3] could be 300x250

Here is an example of index_post.php. The nichemate blue theme was used and copied. I bolded the code.

[adblock3] would be inserted into the post or page itself.
